SYMPTOMS

  • IntelliStart resolution and calibration setup was run
  • Mass positions on tune page and/or acquisition are off more than expected

CAUSE

The Tune page for customers sample was not updated with the correct resolution settings and/or the calibration file was not linked.
These values and file need to be updated after every PM, or after each time the IntelliStart resolution and calibration setup was run.

FIX or WORKAROUND

  1. Update all tune files in use with the following:
    1. Choose the resolution type you want to use (Unit Mass, Quantitative, Low Resolution, or High Resolution)
    2. Link the latest calibration file for the Resolution type selected.

    5. As in the photo above, first clear any previously linked calibration file, and then browse to the most recent file for the resolution type being used. It will be in . . .Calibration file path.PNG
    6. Just select the correct folder for your resolution.
    7. After linking the calibration, you will need to update the LM (Low Mass), HM (High Mass), and ion energies in the ionization mode you are using for both MS1 and MS1 in the tune page. If you are doing a Pos/Neg switching experiment, the settings will need to be set up in both positive and negative modes of the tune page.
    8. You can obtain these values from the report generated after the resolutions are set up by intelliStart.

    10. Take the LM and HM values and update the tune page file.
    11. Note that the Ion Energy is optimized for resolution but not for MRM response. Decreasing the ion energy might lead to decreased MRM response. Either leave the ion energy as is or optimize it. Use one ion in positive and one in negative mode as applicable. Increase ion energy 1 until the peak is no longer increasing. Then do the same for ion energy 2. Ion energy is not compound-dependent but can be different for positive and negative ion modes.
    12. This process must be performed for all tune pages that are in use, otherwise the calibration will not be applied correctly to the system.
