Vion IMS QTof Fluidics Setup warning - WKB993

SYMPTOMS

• Fluidics setup warning in instrument pane and UNIFI logs
• Acquisition stopped
• No lockspray signal seen from reference sprayer
• Lockspray signal with sample sprayer

ENVIRONMENT

• Vion IMS QTof
• UNIFI

CAUSE

• Lockspray beam instability
• Lockspray beam too intense or not intense enough
• Poor mass accuracy

FIX or WORKAROUND

1. Ensure that the lockspray beam is stable:
   1. Purge sample fluidics to remove trapped bubbles.
   2. Check all finger tights are well tighten at the reference valve and pump.
   3. Rebuild lockspray reference probe part number 700004596
   4. Optimize reference probe capillary voltage.
2. Ensure that the lockspray beam is within acceptable limits:
   1. Make up fresh Leucine Enkephalin lockspray solution at 50 pg/ul.
3. Ensure that the lockspray beam is at an acceptable mass window before acquisition:
   1. Check the mass position of Leucine Enkephalin, and adjust if necessary.

ADDITIONAL INFORMATION

Fluidics set up checks that:

• Beam is stable
• Beam can be attenuated to 0.5 IPP and is stable
• Mass accuracy of Leu Enk is within a certain window
