What should be done if TEA or HFIP adducts are detected in nucleic acid analysis? - WKB197867
Article number: 197867
ENVIRONMENT
- LC-MS
- Nucleic acid analysis
ANSWER
Nucleic acids are usually detected as deprotonated ions, but TEA and HFIP adducts contained in the mobile phase may also be detected.
It may be possible to reduce such adducts by changing the TUNE settings.
- Lower the capillary voltage (for TEA adducts).
- Increase the cone gas flow to about 200 L/hr (for HFIP adducts).
- Increase the voltage of the sampling cone (If it is raised too much, some nucleic acid ions may be cleaved).
ADDITIONAL INFORMATION
If this does not help, try repreparing the mobile phase, cleaning the instrument, and continuously running blank sample.
