ACQUITY Arc: What is the Stroke Volume and Plunger Wash and what parameters should be modified when switching between UPLC/HPLC modes? - WKB92823

Article number: 92823

ENVIRONMENT

  • ACQUITY Arc

ANSWER

■Stroke Volume
The default stroke is 132μL, but when the movement of the plunger (Stroke Volume) is large and the baseline fluctuates due to mixing,
it can be expected that the baseline fluctuation will be reduced by setting a small stroke volume. However, if it is set small, the maximum flow rate will also be small.

■Plunger Wash
Clean the plunger with seal wash solvent. While cleaning the seal, fill the pump with the current solvent composition and push it out.
This is recommended to be performed after using a buffer solvent

■Parameters to be modified when switching between HPLC/UPLC modes
- Considerations about the sampling rate of the detector
A minimum of 15 points is required from the peak start to peak end. And this is calculated from the width of the narrowest peak detected, in seconds.
Set to 1 or 2 points/sec for HPLC mode
Set to 10 or 20 points/sec for UPLC mode
.

- Setting Pass 1 and 2
Select the path according to the system volume of the method transfer source. Gradient delay varies depending on the system volume.
Pass 1 1100uL
Pass 2 700uL

Press the F1 key while the Instrument Method is open with Empower, the Help window for the corresponding item appears.
