How to clean the LC flow path? - WKB85249

Article number: 85249

OBJECTIVE or GOAL

  • Clean the LC flow path

ENVIRONMENT

  • ACQUITY UPLC H-Class
  • ACQUITY Arc
  • Alliance HPLC

PROCEDURE

1. Preparation of the solution
Prepare a solvent prepared by adding 0.1% formic acid to water : acetonitrile : methanol : isopropanol = 1:1:1:1.
Use this solution for flow path cleaning and injector cleaning. Prepare one vial containing the above cleaning solution for injector cleaning.

2. Place the A-D line and the purge line into the solution from step 1.

3. Prime all lines and the purge line. (In case of Alliance2695, perform the WetPrime and PurgeInjector.)

4. Remove the column and connect the union. If there is a detector such as PDA, route its outlet to the waste.
(Ensure that the flow is not going to the QDa.)

5. Create an instrument method with a flow rate of 0.5 mL/min with A:B:C:D=25:25:25:25 and register it in the method set.

6. Create a sample set. Set the run time to 10 minutes, put the line in the vial from step 1,
and create a sample set to make 30 injections with the maximum injection volume (50 μL when there is an expansion loop, otherwise 10 μL, 100 μL for Alliance 2695), run the sample set.

7. After cleaning, prime the entire instrument with water and then switch to the normal mobile phase.
