How to flush an ACQUITY Premier - WKB269769

OBJECTIVE or GOAL

Flush or clean the ACQUITY Premier System within the limits of High Performance Surfaces. (Order the LC Purging Kit (186010210) to perform this procedure.

PROCEDURE

1. Remove the column  and put a union in its place.
2. Remove the solvent resevoir sinker / filters.
3. Plumb the outlet of the union to waste, not the MS (optionally, use a back-pressure line).
4. BASE Flush: Place all lines in a 0.1% aqueous ammonia solution in water.
5. BSM / QSM : Prime all lines for 5 minutes each. FTN: 60s Wash, 30 cycles on syringe. -or- FL: 15 cycles
6. Run the BASE solution through the lines at 0.2 mL/min for 20 min (this can be extended if needed).
7. Place all lines in fresh, 100% water (LC-MS grade/MilliQ).
8. BSM / QSM: Prime all lines for 5 minutes each. FTN: 60s Wash, 30 cycles on syringe. -or- FL: 15 cycles
9. Run the water through the system at 0.2 mL/min until the pressure is stable.
10. Place all lines in the IPA Flush: 95% IPA /5% water
11. BSM / QSM: Prime all lines for 5 minutes each. FTN: 60s Wash, 30 cycles on syringe. -or- FL: 15 cycles
12. Run the IPA solution through the system at 0.1 mL/min for 20 min (this can be extended).
13. Optional: If the column is compatible with the ACID Flush, connect the column so that it can also be flushed.
14. ACID Flush: Place all lines in ACN:IPA:MeOH:H2O and 1% formic acid.
15. SM: Prime all lines for 5 minutes each. FTN: 60s Wash, 30 cycles on syringe. -or- FL: 15 cycles
16. Run the ACID solution through the system at 0.2 mL/min for 20 min (this can be extended).
17. Make up fresh mobile phase and connect the column to the MS.
18. BSM / QSM: Prime all lines for 5 minutes each. FTN: 60s Wash, 30 cycles on syringe. -or- FL: 15 cycles
