The operator's guide includes the following tasks, which can help improve chromatography and sensitivity issues:

1. Perform HPLC system passivation.

2. Clean the working electrode.

3. Maintain the reference electrode.

4. Clean the HPLC column per the manufacturer's care and use guide.

5. Use only HPLC-grade water.

6. Use the correct flow cell spacer. If any maintenance requires you to disassemble the flow cell, the spacer must be replaced with a new one. It shouldn’t be reused.

7. If the HPLC system is idle for more than 48 hours, remove any buffers from the system and flush with water. Use 50:50 water/methanol for storage.

Returning the system back to buffers calls for the reverse—prime with water and then with buffers.

8. If you are not using your HPLC column for a similar period, it should be flushed following the manufacturer's care and use guide—typically, flush off buffers with water and then store in water/methanol. If you are changing the column, note the requirement to flush at low flow for 3 days.

9. A periodic flow cell flush is beneficial—30 minutes at 0.5 mL/min with water or ethanol with column removed.

10. To prevent formation of air bubbles in the flow cell, the flow cell should be positioned at a 45-degree angle, and the waste line outlet tubing from the flow cell should run to a waste container that is at a higher level than the flow cell. This gives a minute back pressure that prevents air-bubble entrapment.

The outlet tubing should be under the liquid level to prevent electrical baseline noise.
