UNIFI Glycan Assay (FLR with DDA confirmation) assigns the wrong glycan mass - WKB77488

Article number: 77488

SYMPTOMS

  • Glycan Assay (FLR with DDA confirmation) assigns the wrong glycan mass to an observed peak

ENVIRONMENT

  • UNIFI 1.9.4

CAUSE

The Glycan Assay (FLR with DDA confirmation) analysis method carries out glycan assignments using the FLR data and glycan units (GU) read from the GU standard curve. The DDA data is not used directly in the UNIFI analysis. In order to confirm the FLR GU identification using the collected DDA data, you must export the data to Simglycan.

If the GU match is within the specified limits, the FLR peak is matched to a particular glycan, even if the mass appears to be outside of the expected ppm error limits.

FIX or WORKAROUND

  1. Export the DDA data as a LCS file and analyze in Simglycan.
  2. If alternative assignments are available that match both the GU result and the expected mass, reassign the identification manually.
