Will an 8-µL injection that contains 50% chloroform harm my ACQUITY UPLC CSH C18 column? - WKB98522

Article number: 98522

ENVIRONMENT

  • UltraPerformance Liquid Chromatography (UPLC)
  • Solvent compatibility
  • Charged surface hybrid (CSH)
  • ACQUITY UPLC CSH C18

ANSWER

No, chloroform injected onto the column will not harm the packing.

ADDITIONAL INFORMATION

Caution must me used to ensure that chloroform injected onto the column is fully compatible with the mobile phase.

For example, if a sample containing chloroform is injected into an aqueous mobile phase, an emulsion can form because chloroform and water are not miscible.

It this happens, flush the column with 10 column volumes of water (to remove buffer), followed by 10 column volumes of 100% acetonitrile or methanol.

If no buffer is used, simply flush the column with 10 column volumes of of 100% acetonitrile or methanol to wash off residual chloroform.
