How do I convert a Waters Spherisorb Amino (NH2) normal-phase column to run a reverse-phase or HILIC method? - WKB102321
OBJECTIVE or GOAL
Switch solvents from normal phase to reverse phase or HILIC.
ENVIRONMENT
- NH2
- Solvent switching
- Hydrophilic interaction chromatography (HILIC)
- Solvent changeover
PROCEDURE
1. Flush with 10 column volumes of 100% isopropanol.
2. Flush with 10 column volumes of 100% Acetonitrile to reduce backpressure when moving into next step
3. Flush with miscible aqueous-organic mobile phase with no buffer for 50 column volumes. (Example: 50:50 water/acetonitrile)
4. Flush with initial mobile phase with buffer (if using buffer) for 20 column volumes.
ADDITIONAL INFORMATION
Waters Spherisorb columns ship in 96:4 heptane/isopropanol.
