HDX: How to desalt from TCEP contamination - WKB5266

Article number: 5266

OBJECTIVE or GOAL

  • Tris-2-chloroethyl phosphate (TCEP) is known to give high background noise.

  • The following suggestion will better maintain the system and minimize salt accumulations.

ENVIRONMENT

HDX systems

PROCEDURE

Before proceeding, everything must be cleaned including sonication of injection valve and replacement tubing.

  1. Keep TECP concentration low. Use 0.5 M TCEP in quench buffer, which means 0.25 M in final solution after mixing with labeled sample.
  2. Run a clean blank after every sample run.
  3. Keeping desalting flow rate at 200 ul/min at 4 min is a good way to maintain the system and minimize the salt accumulation.
  4. Acquire MS from 300 Da starting mass to work around the problems created by the 251 peak.
