HDX: How to desalt from TCEP contamination - WKB5266
Article number: 5266
OBJECTIVE or GOAL
-
Tris-2-chloroethyl phosphate (TCEP) is known to give high background noise.
-
The following suggestion will better maintain the system and minimize salt accumulations.
ENVIRONMENT
HDX systems
PROCEDURE
Before proceeding, everything must be cleaned including sonication of injection valve and replacement tubing.
- Keep TECP concentration low. Use 0.5 M TCEP in quench buffer, which means 0.25 M in final solution after mixing with labeled sample.
- Run a clean blank after every sample run.
- Keeping desalting flow rate at 200 ul/min at 4 min is a good way to maintain the system and minimize the salt accumulation.
- Acquire MS from 300 Da starting mass to work around the problems created by the 251 peak.
