How to improve high mass transmission on SYNAPT G2-Si MALDI using the quad profile - WKB4952

OBJECTIVE or GOAL

Adjust the manual setting of the quad profile to bias high mass transmission and remove matrix noise.

ENVIRONMENT

  • MALDI
  • SYNAPT G2-Si

PROCEDURE

  1. Click the Setup drop-down in the tune page.
  2. Open the Quad Profile menu.
  3. Select Manual Profile and use the following:
    1. 2000    4   1
    2. 7000   90  5
    3. 7000

ADDITIONAL INFORMATION

Note: Observing ions at 5000 m/z is routine for an 8k quad, and up to 16,000 m/z is possible with focused tuning (the high mass example data from the service note shows the Myoglobin peak).

For a 32k quad, there is little change in sensitivity over the mass scale, but you have increased selectivity.
