How to calibrate the LCT Premier XE using the calibration wizard - WKB32438
Article number: 32438
OBJECTIVE or GOAL
To successfully calibrate the LCT Premier or LCT Premier XE
ENVIRONMENT
- LCT Premier
- LCT Premier XE
PROCEDURE
- Infuse sodium formate solution into the analyte probe and ensure that a spectrum of sodium formate is observed on the MS Tune page.
- Run the Calibration Wizard, selecting a mass range of 50 to 1000 Da, and the reference file NaFormatePosES.ref.
- Ensure that a calibration curve with a RMS residual of less than <5e-04 amu (or less than 3 ppm) is produced.
- 4. If required, click Edit > Calibration Parameters and vary the intensity threshold to remove any small peaks from consideration by the software.
NOTE: If required, the polynomial value can be adjusted to achieve a better mathematical fit for the calibration curve. - Outlier peaks that are affecting the calibration fit (such as those of insufficient intensity) can be removed from the calibration.
- 6. If required, left-click and drag to zoom in on the peak to be removed, then right-click on the peak in the middle trace, then right-click on the peak in the top trace to remove it from the calibration calculation.
NOTE: The removal of too many peaks results in a suspect calibration calculation. - Click Finished > Accept Calibration… once optimization is complete to accept the calibration.
- Click Finish to complete the Wizard.
