OBJECTIVE or GOAL

Clean the ACQUITY UPLC H-Class and I-Class Systems

ENVIRONMENT

  • ACQUITY UPLC (Classic)
  • ACQUITY UPLC H-Class
  • ACQUITY UPLC H-Class Bio
  • ACQUITY UPLC H-Class PLUS
  • ACQUITY UPLC H-Class PLUS Bio
  • ACQUITY UPLC I-Class

PROCEDURE

  1. Prepare the washing solvent (see below)
  2. Prepare the washing solvent in a sample vial and place in autosampler.
  3. Remove columns
  4. Remove the solvent sinker / filters from the solvent manager lines.
  5. Prime all lines (solvent manager and sample manager) except seal washing line with the washing solvent for 5 minutes.
  6. Create the a new method to be used for cleaning
    A/B/C/D=25/25/25/25 Flow rate: 0.2mL/min
  7. Create a new  sample list / sample set to be used for cleaning
Injection Volume: Needle (including the expansion loop) volume
    Run Time: 1 minute
Number of Injections: 30 to 100
  8. Rinse all the lines with Methanol before going back to the original solvents used in the method.

ADDITIONAL INFORMATION

For the washing solvent, select one which can dissolve the samples analyzed routinely.
If you can't find the appropriate solvent, use 1:1:1:1 water / acetonitrile / methanol / isopropanol with 0.2% formic acid.
If alkaline solution is expected to be more effective, 0.2% formic acid can be replaced with 0.2% aqueous ammonia.
In either case, after cleaning, replace the washing solvent with water and perform the above cleaning procedures to switch to water thoroughly.
Even after completing 30 injections, it is more effective to keep the washing solution running if possible.
To give a rough idea, prepare 1L of washing solvent and use it all.
If you want to shorten the time, perform the cleaning at a higher flow rate.
