How to calibrate an ACQUITY UPLC FLR Detector - WKB56294

OBJECTIVE or GOAL

Follow the protocol for calibrating the ACQUITY UPLC FLR module.

Two parameters must be calibrated within this optical detector before using the instrument:

  • The fluorescence wavelength (Excitation and Emission)
  • The PMT (PhotoMultiplier Tube) sensitivity response

PROCEDURE

Note that the lamp must be lit for at least an hour before starting the calibration processes.

To calibrate the ACQUITY UPLC FLR, run 100% water through the system. [Ensure that the water is clean. Request MS-grade solvent when working with mass spectrometers.]
In Service or Maintenance mode, perform the following setup in the exact order shown:
- Calibrate wavelength (approx. 2 minutes)
- PMT Calibration (approx. 2 minutes; a pass is for R > or = 0.999)
- Normalize units (approx. a few seconds). RAMAN gain would be about 200 for new modules or modules with a new lamp fitted.

ADDITIONAL INFORMATION

In a UNIFI environment, if a request to select a calibration curve appears after completion of calibration procedures but before a successful run of the FLR precision test (see screenshot),

click "Cancel" and then "Forward". The request clears.
