How can we address the tailing effects seen with protein-based SEC and IEX methods? - WKB231489

ANSWER

Tailing is a known effect seen with the interaction of proteins with the analytical Teflon light-guided cell when run with high salts (such as those used in SEC and IEX methods).

The stainless steel flow cell and titanium flow cell are usually suggested, although some tailing may still be observed with the stainless steel flow cell. This is due to the nonspecific metal chelation of proteins to stainless steel surfaces. - WKB230675

This is why the titanium cell is recommended for SEC and IEX separations. Refer to the Guidelines for Routine Use and Maintenance of Ultra Performance SEC and IEX separations, where it is shown that the use of a 5-mm titanium flow cell improves the peak shape.
