What are the guidelines for RI detector purge time when using a new column and mobile phase? - WKB224716

Article number: 224716

ENVIRONMENT

  • Alliance HPLC System
  • 2695 Separations Module
  • Arc HPLC System
  • ACQUITY Arc
  • ACQUITY UPLC
  • ACQUITY UPLC H-Class
  • 2414 Refractive Index Detector
  • 2998 Photodiode Array Detector
  • ACQUITY UPLC RI Detector

ANSWER

  • Perform a RI detector purge after passing at least 20 times the column capacity through the column.
  • Incomplete column equilibration can affect the chromatogram, even with purging.
  • When using a new column or changing the composition of the mobile phase, be sure to fully equilibrate the column.
  • Depending on the type of column and mobile phase, it may be necessary to pass 20 times or more of the column capacity.
  • Purge the RI detector for at least 30 minutes.
