How do I use Helium sparge with preparative chromatography? - WKB11501

Article number: 11501

OBJECTIVE or GOAL

To show an approach to setting up Helium sparge for inline mixing of two mobile phases.

ENVIRONMENT

2535/2545/2555 Quaternary Gradient Module

PROCEDURE

  1. Obtain and secure a tank of 0.9990 purity Helium with a 2-stage regulator.
  2. Connect a stainless steel line with 0.040 ID from the regulator to the bulkhead fitting on the pump.
  3. Use three hole caps (WAT062479) for 1 L bottles or (WAT062341) for 4 L bottles.
  4. Place the solvent and sparge line through the cap and into the mobile phase bottle.
  5. Place a solvent filter upside-down on top of the cap through the third hole.
  6. Set second stage of the regulator to 100 psi.
  7. Turn the sparge on to 100% flow for 20 minutes for every 1 L of mobile phase.
  8. With desired mobile phase flow set, adjust Helium flow to 20%.

ADDITIONAL INFORMATION

You can use this procedure for all pumps without an inline degasser.
