How to extract and integrate the C12 and C13 peaks for a compound in UNIFI - WKB7395

OBJECTIVE or GOAL

Integrate the parent (C12) chromatographic peak and the first isotope (C13) chromatographic peak in UNIFI.

PROCEDURE

  1. In the Analysis, go to the Investigate tab and select processing options in the top-right.
  2. Pull up the Chromatogram of the sample to be analyzed. 
  3. Under Operations, select "Extract mass chromatogram" and input the mass range of the parent or C12 ion. Use a 10-ppm or smaller window and click the start button at the bottom. This extracts the chromatogram of the C12 ion.
  4. Select Integrate from the Operations drop-down menu. 
  5. Right-click on the Integrated peak and Select properties.
  6. Go to Annotations and Peak Annotations in the Chromatogram Properties window.
  7. Expand Integrated LC System - Suitability Peak, scroll down to Area, and then select the blue down arrow to add it to Peak information.
  8. Select OK.
  9. The peak's area is now listed in the Chromatogram window for those peaks integrated.
  10. Repeat the extract mass chromatogram and integration for the C13 mass.
