In UNIFI, the same component identified in several samples is split into two component summary charts - WKB21021

SYMPTOMS

  • UNIFI identified the same component in several or all samples. However, the component summary graph/table does not report the component for all samples in a single graph. Instead, the results for the same identified component are split into two or more summary graphs. Example:

CAUSE

The retention time window specified in Binary Sample Compare is too small.

UNIFI takes the center of the retention time (RT) window used for sample-to-sample component matching from the RT of the identified component in the reference sample. If there is a lot of RT variability from sample to sample and the retention time window in the Binary Compare settings is not wide enough to allow UNIFI to match all the samples to the reference sample, it may identify the same component in all the samples but not match them to the reference sample. In that case, two or more component summary charts may be created, one for those that lie within the RT window either side of the reference sample RT and another for those outside of the RT window.

FIX or WORKAROUND

  1. Increase the retention time window parameters in the Binary Sample Compare settings of the processing method (see below).

  1. Reprocess the dataset.
