Processing peptide mapping data uses all memory - WKB108869
Article number: 108869
SYMPTOMS
- Peptide mapping that contains numerous disulfide bonds and missed cleavages can generate many
fragment ions, which results in a memory error
ENVIRONMENT
- UNIFI 1.9.4

CAUSE
- CRI-138 (INFULA-1201)
FIX or WORKAROUND
Create a map of potential disulfide bonded regions of a protein by performing a mass
spectrometry search with and without disulfide bonds. Then, add one or two disulfides to the
protein and gradually add the information to the peptide map.
For complex disulfide patterns with more than two disulfides per peptide, consider removing peptide chains from the sequence, and then create a custom Cys modification of that chain, using a look ahead and/or look behind sequence in the modifier to direct that chain to a specific Cys residue.
