OBJECTIVE or GOAL

To view the ratio of the analyte area to the internal standard (IS) area in the TargetLynx browser.

PROCEDURE

Use the peak response field. 

peak response = analyte area / IS area x IS concentration

When the concentration of the internal standard (IS) is set to fixed 1.0:

peak response = analyte area / IS area

  1. Select the internal standard in the left pane of the TargetLynx method.
  2. Click Compound properties.
  3. Set the Response Type to External (absolute - no internal standards).
  4. Click Calibration properties.
  5. Set Propagate Calibration Parameters? to No.
  6. Set Concentration of Standard: level to Fixed.
  7. Set Concentration of Standard to 1.0.
  8. Select the compound in the left pane.
  9. Click Compound properties.
  10. Set Response Type to Internal (relative - use internal standards).
  11. Select the internal standard in the Internal Standard: 1 field.
  12. Save the method.
  13. Process samples.
  14. Right-click in the summary table, and then select Change Column Order.
  15. Highlight Peak Response in the left pane and click Add.
  16. Click OK to close. The response column appears in the summary table.
  17. Select File > Save Layout As to save the browser layout.
