Manually tune compounds to optimize signal.

PROCEDURE

  1. Open the MS Tune page for the relevant method.
  2. Ensure that the most recent calibration is linked and that the Low Mass/High Mass Resolution values reflect the calibration.
  3. Tune mass positions using the appropriate tuning solution.
    Manually tune using combined mode by infusing the solution and flowing at a low rate (0.1-0.2 mL/min), preferably with a gradient similar to that where the analyte comes out.
  4. Tune capillary voltage while in combined mode, to mimic full LC-MS sample analysis.
  5. Tune cone and collision voltages to ensure optimum performance for the compounds.
  6. Adjust Ion Energy 1 and 2 to maximize the signal for the most difficult compound. If applicable, also adjust the collision gas flow to enhance signal.
  7. Adjust temperatures and gas flows to accommodate the LC flow.
  8. Make note of any changes, and update the MS method with the relevant parameters. If mass positions have changed, also update the processing method.
