Manually tune compounds to optimize signal.

PROCEDURE
- Open the MS Tune page for the relevant method.
- Ensure that the most recent calibration is linked and that the Low Mass/High Mass Resolution values reflect the calibration.
- Tune mass positions using the appropriate tuning solution.
Manually tune using combined mode by infusing the solution and flowing at a low rate (0.1-0.2 mL/min), preferably with a gradient similar to that where the analyte comes out. - Tune capillary voltage while in combined mode, to mimic full LC-MS sample analysis.
- Tune cone and collision voltages to ensure optimum performance for the compounds.
- Adjust Ion Energy 1 and 2 to maximize the signal for the most difficult compound. If applicable, also adjust the collision gas flow to enhance signal.
- Adjust temperatures and gas flows to accommodate the LC flow.
- Make note of any changes, and update the MS method with the relevant parameters. If mass positions have changed, also update the processing method.
