When using an internal standard, Empower takes the response (Y value) of the component and divides it by the response of the internal standard. It then takes this result and multiplies it by the amount of the internal standard. This becomes the Y value for the component, which is then plotted against the X value of the component.
ADDITIONAL INFORMATION
Here is the equation: (Y value of component/Y value of IS)*(X value of IS). This creates a point on the calibration curve for the component in question. Because the amount of the internal standard is being used to generate the curve, you must include the internal standard and its amount in the unknown samples injections. This will allow for accurate quantitation.
