The pressure required is different throughout the SPE protocol and varies depending on the viscosity of the samples.

Load and wash steps might require lower pressure (roughly 5 psi) that would need to be adjusted to reach the target flow rate of one drop per second. The pressure at the elution step would be set higher to remove all the liquid from each well, and this will also vary depending on the vacuum pump system and the number of samples to be processed at one time.
