What do I need to consider, if I am running a buffer outside its' buffering range? - WKB73783

Article number: 73783

ENVIRONMENT

  • Buffer
  • buffer capacity
  • pH
  • range
  • mobile phase
  • method considerations

ANSWER

If the method mobile phase pH is outside the buffering range of a buffer, consider the following to minimize variation in the chromatography:

1) Make up the buffer more often to maintain the optimal ion concentation, especially if the buffer is volatile

2) Increase the concentration of the buffer so there are more ions in solution

3) Control of the pH at preparation and from daily drift in a container, especially if the buffer is volatile
