How do I minimize peak splitting problems when running HILIC mode chromatography? - WKB80284

Article number: 80284

SYMPTOMS

Peak Splitting

ENVIRONMENT

  • column
  • HILIC
  • Amide
  • ACQUITY
  • XBridge
  • UPLC
  • HPLC
  • polar retention
  • peak split
  • peak shape
  • troubleshooting

CAUSE

Too much water in the sample diluent.

FIX or WORKAROUND

  1. Dilute the sample in 50/50 acetonitrile/water.
  2. Reduce the injection volume. This is determined experimentally.

ADDITIONAL INFORMATION

Water is the strong solvent in HILIC mode separation. Too much highly polar solvent can produce a strong solvent effect and influence the peak shape when injected into the weaker solvent conditions. The weaker solvent in HILIC mode separation is the acetonitrile.
