ANSWER

Before you use a new BEH HILIC column, you must flush it with 50/50 Acetonitrile:Water with 10mM of the buffer used in the method for 50 column volumes.

After the 50/50 Acetonitrile:Water with 10mM final buffer step, equilibrate the column for 20 column volumes in the initial conditions of a gradient or the isocratic mobile phase.

Note: If you are using a BEH HILIC column with low concentrations of mobile phase additives, you may need 100- 200 column volumes for complete equilibration. In addition, mobile phases that contain formate may also require a longer initial column equilibration.
