How do I calculate the empty column volume of a column? - WKB48971

Article number: 48971

OBJECTIVE or GOAL

Determine the volume of an empty column.

ENVIRONMENT

  • Column
  • calculate
  • empty volume
  • HPLC
  • UPLC
  • ACQUITY
  • HSS
  • Symmetry
  • BEH
  • OST
  • APC
  • XBridge

PROCEDURE

  1. Use Volume = pi x radius2 x Length
  2. pi = 3.14
  3. r2 and Length should be converted to centimeters
  4. Diameter of column divided by 2 = radius
  5. radius x radius = r2
  6. 3.14 x  r2  x L = Volume in cm3
  7. cm3 = 1 mL

ADDITIONAL INFORMATION

The empty column volume of 4.6 mm x 150 mm is 2.5 mL.

pi = 3.14

4.6 mm  becomes 0.46 cm and 150 mm becomes 15 cm.

0.46 cm divided by 2 =  0.23 cm radius

0.23 cm x 0.23 cm = 0.0529 cm2

3.14 x 0.0529 cm2 x 15.0 cm = 2.49cm3
