How to clean a µBondapak C18 column - WKB252273

OBJECTIVE or GOAL

Clean a µBondapak C18 column.

ENVIRONMENT

PROCEDURE

1. Remove the guard column if one is in use.
2. Remove the pre-column filter if one is in use.
3. Disconnect the column outlet from your detector.
4. Flow the outlet of the column to waste (bypass detector).
5. Prepare a cleaning solution of 50:50 dimethylsulfoxide/water.
6. Run a mobile phase of 100% water.
7. Make repeated injections of the 50:50 dimethylsulfoxide/water cleaning solution every five minutes.
8. Repeat step 7 until ten empty column volumes of mobile phase have passed through the column.
9. After the cleaning injections are made:
   • Flush the column with 10 empty column volumes of water.
   • Flush the column with 10 empty column volumes of methanol.
   • Flush the column with 10 empty column volumes of chloroform.
   • Flush the column with 10 empty column volumes of methanol.
   • Flush the column with 10 empty column volumes of water.
