ANSWER

The void volume in GPC and SEC is the exclusion volume of the column.

Vo = exclusion volume. Use approximately 0.4 x empty SEC/GPC column volume.

Vt = total retention volume (words used in Empower GPC software parameter) [totally included volume]. Use approximately 0.8 x empty SEC/GPC column volume.

ADDITIONAL INFORMATION

Calibration of actual molecular weight standards is required to establish the actual exclusion volume and included volume for the samples, because their size and shape are affected by the method conditions, which can include sample/standard diluent and mobile phase.
