ANSWER
Flush the ACQUITY BEH HILIC column for 20 column volumes with 100% acetonitrile to remove the water; follow with 100% iso-propanol for 20 column volumes. Then follow with the normal phase solvent for 50 to 200 column volumes to equilibrate the column.
ADDITIONAL INFORMATION
Use of 100% acetonitrile prior to 100% IPA will lower the back pressure of the mixing solvents because most of the water will be gone by the time the IPA is introduced.
