Can I clean an XBridge or ACQUITY BEH SEC column with guanidine or urea? - WKB21234

Article number: 21234

ENVIRONMENT

  • ACQUITY UPLC
  • ACQUITY BEH SEC
  • XBridge BEH SEC
  • column
  • clean
  • urea
  • guanidine

ANSWER

Yes. Flush the column with 10 column volumes of UPLC-grade water (for ACQUITY UPLC) or HPLC-grade water (for HPLC) to remove buffers.

Guanidine: Prepare a cleaning solution of up to 6M guanidine HCl.

Urea: Prepare a cleaning solution of up to 8M urea. Adjust the pH of this solution to pH 7.0 with phosphoric acid.

ADDITIONAL INFORMATION

Either urea or guanidine HCl can be used, but the pH of these solutions should be buffered and adjusted to a pH of 7.0 to ensure that the cleaning solutions are compatible with the pH limits of the column that is being cleaned.

After using the cleaning solutions, sufficient cleaning with UPLC or HPLC grade water will ensure the urea and guanidine have been washed from the column.
