How should I clean a CORTECS C18 column if I suspect protein has precipitated? - WKB205034

Article number: 205034

ENVIRONMENT

  • Column care and use
  • Column cleaning
  • CORTECS C18+
  • DMSO
  • Proteins

ANSWER

Option 1: Inject repeated aliquots of dimethyl sulfoxide (DMSO). With DMSO, the recommended cleaning injection solution should be 50:50 DMSO/H2O in a sample vial. Then, run 100% H2O as mobile phase and make 5 to 10 injections of the 50:50 DMSO/H2O solution as the cleaning solution.

Option 2: Gradient of 10% to 90% B where A = 0.1% trifluoroacetic acid (TFA) in water and B = 0.15% trifluoroacetic acid (TFA) in acetonitrile.

Option 3: Flush column with 7M guanidine hydrochloride or 7M urea.
