How much protein can I load onto an Ultrahydrogel column if I elute it with 50% acetonitrile? - WKB126058

Article number: 126058

ENVIRONMENT

  • High Performance Liquid Chromatography (HPLC)
  • UltraPerformance Liquid Chromatography (UPLC)
  • Gel permeation chromatography (GPC)
  • Size exclusion chromatography (SEC)
  • Gel filtration chromatography (GFC)
  • Ultrahydrogel
  • XBridge
  • Bridged ethylene hybrid (BEH)
  • Protein separation
  • ACQUITY

ANSWER

This would need to be determined experimentally.

Note that elution with 50% acetonitrile is not recommended because proteins will precipitate on the column in 50% acetonitrile.

ADDITIONAL INFORMATION

Ultrahydrogel columns are not designed to capture proteins via nonspecific adsorption.

Waters cannot guarantee any type of reproducible results when relying on nonspecific binding as a way to separate proteins from small molecules on Ultrahydrogel columns. Due to the charged surface of Ultrahydrogel packings, protein separation is not recommended on these columns.

Instead, diol-bonded SEC columns, such as BEH SEC columns, are recommended because proteins are not as likely to adsorb to these packings via nonspecific adsorption.
