How do I clean my µBondapak Cyano (CN) column? - WKB123612

OBJECTIVE or GOAL

Clean a µBondapak Cyano column.

• Column care and cleaning

PROCEDURE

1. Remove the guard column.
2. Prepare a solution of 50:50 dimethylsulfoxide (DMSO)/water.
3. Disconnect the column outlet from the detector and catch flow in a beaker.
4. Run 100% water as the mobile phase.
5. Make 200-µL injections every 5 minutes until a total of 10 column volumes of water has passed through the column.
6. After cleaning, connect the guard.
7. Connect the column to the detector.
8. Equilibrate the column with working mobile phase.
9. Make an injection to check performance.

ADDITIONAL INFORMATION

If the cleaning above does not correct the issue, you can try a column regeneration procedure:

1. Remove the guard column.
2. Disconnect the column outlet from the detector and catch flow in a beaker.
3. Run 10 column volumes of each of the following solvents through the column:
   • Water
   • Methanol
   • Chloroform
   • Methanol
   • Water

Note: The formula to calculate your column volume is:  pi * r² * L   (r = radius in cm, L = length in cm)
