How do I reduce the back pressure in a method that is being developed at a high flow rate? - WKB118861

Article number: 118861

OBJECTIVE or GOAL

Reduce the back pressure of an LC method and stay within the column pressure limits.

ENVIRONMENT

  • Method development
  • Column selection

PROCEDURE

In order to run a column within its manufacturer's suggested pressure ranges, one or several of the following options would be implemented.

  • Use a shorter column length.
  • Use a wider column ID.
  • Use a larger particle size.
  • Raise the column temperature.
  • Reduce the flow rate
  • Consider using larger ID tubing post-column to reduce the friction produced by the method running at very high flow rates.
