Reduced digestion efficiency on new Enzymate BEH Pepsin column attached to HDX system - WKB117306

SYMPTOMS

• New Enzymate BEH Pepsin column is not digesting Phos B standard efficiently and is giving very low recovery of expected peptides at startup

ENVIRONMENT

• HDX system
• Enzymate BEH Pepsin column
• HDX Phosphorylase B standard

CAUSE

Quenching buffer reagents are going bad.

FIX or WORKAROUND

1. Check Enzymate column efficiency with the Waters HDX Phosphorylase B standard {186006930) and compare peptide digestion recovery.  
2. Quench labeling reaction with an alternative buffer.
3. Remake method quenching buffer from a fresh lot of reagents.
