The Rapid Buffer sold in each kit is considered a "5X" concentrated buffer. Waters does not sell this as a "5X" buffer outside of the GlycoWorks RapiFluor-MS N-Glycan Kit.

However, Waters does sell 186008100 - Glycoworks Rapid Buffer, 5 mL, which contains 5.0 mL of 50 mM HEPES pH 7.9. This is considered a "1X" concentration. It can be used in a modified protocol where this 1X Rapid Buffer is used to replace the water (for sample dilution) in the standard deglycosylation step.

Examples:

  • Standard protocol (with 5x buffer): 7.5 µL of 2 mg/mL protein + 6 µL of RapiGest (5x buffer) +15.3 µL of water
  • Modified protocol (with 1x buffer):  7.5 µLof 2mg/mL protein (dilute/buffer exchange with the 1x buffer) + 6 µL of RapiGest (1x buffer) +15.3 µL of 1x buffer.

In both cases, the pH should be stable at about 7.9. There is no need to check the pH or re-titrate.
