The purpose of derivatizing amino acids is to attach a tag that will allow for retention of the polar and nonpolar amino acids and separation in reverse phase chromatography—also, to tag the amino acids so they can be detected and quantitated.
Many amino acid structures are not detectable by Fluorescence or UV without derivatization. They are in low abundance and would not be detected by optical detection systems without derivatization.
