PROCEDURE

  1. Power-on the workstation and log in to Windows. Do not start MassLynx at this time.
  2. Power-on the component of the UPLC system that has the network switch. This is generally the Sample Manager. On older systems it may be the Column Manager. This component will have multiple network cables connected on the back. The other components will each have only one network cable. Allow the component 30 seconds to 1 minute to initialize. 
    Caveat: if your system has the Sample Organizer, this must be powered-on before powering-on the Sample Manager. You only need to wait about five to ten seconds between powering-on the Sample Organizer and powering-on the Sample Manager.
  3. Power-on each additional component of the UPLC system, one at a time, waiting 30 seconds to 1 minute between each. The order does not matter here. 
  4. When all UPLC components have been powered-on, turn on the power to the mass spectrometer. Wait five minutes for the mass spectrometer to initialize. The mass spec takes longer that the UPLC components because it has a computer embedded in its electronics that needs to boot up, and the system is far more complex that the UPLC. 
  5. After five minutes, open MassLynx. Wait for the software to completely open, initialize, and connect to the instruments. Along the bottom edge of the sample list page, near the right-hand corner, look for the MS status to say Not Scanning. 
  6. Open the MS Tune page. From the menus, go to Vacuum > Pump. This will start the vacuum system. Note that the vacuum system will not start automatically when the instrument is powered-on. 
  7. To achieve a stable vacuum, allow the vacuum system to pump 18 to 24 hours before running samples.
