ANSWER

It is suggested that water be in the flow cell when performing the manual calibration.

 

ADDITIONAL INFORMATION

Other clear solvents can be used, but you do not want to use anything with a high background that could interfere with the recognition of the erbium peaks. If water is running through the cell, then wavelength calibration, PMT calibration, and normalization can be run in succession without changing mobile phase.
