Poor chromatographic peak shapes on Acquity - WKB54306

SYMPTOMS

  • Poor chromatography - Peak Tailing
  • Failing QC samples
  • Decreasing peak response
  • Leak test fails with hex cap on column inlet fitting
  • Reproducibility issues

ENVIRONMENT

  • ACQUITY UPLC I-Class (Plus and Bio)
  • ACQUITY UPLC H-Class (Plus and Bio)
  • ACQUITY UPLC Column Manager
  • ACQUITY UPLC Column Heater

CAUSE

  • Void in flow path
  • An improperly fitted column in the column manager

Finger-tight column fittings that were tightened using tools may be damaged. The two-piece metal ferrule between the finger-tight fittings may have been pushed out of position.

  • Improperly fitted tubing/loop in the flow path

FIX or WORKAROUND

  1. Refit the column/tubing/loop
  2. Replace the APH or post-column tubing if it is damaged due to over-tightening with tools. (205001757 - ACQUITY I-Class APH, SS, 12.5LG - PLUS and 700008868 - ASSY, WELDED TUBE, SS, .003 ID X 19.0 LG)

ADDITIONAL INFORMATION

PEEK ferrules should be replaced after every ten to twenty column changes.

APH of a different length may be required for some column manager configurations.
