With 2695 Separations Module and 2489 UV/Vis Detector, area values are much lower than before - WKB238825

Article number: 238825

SYMPTOMS

  • Only one peak detected
  • The shape and retention time are the same as usual, but the height is five times lower
  • Performed three injector purges and one compression check
  • Measured with this device once and the area was low, then moved the column, sample, and mobile phase to another 2695 purchased around the same time and the run finished with no problems
The next day, moved the same column, sample, and mobile phases back to this device and the area value was still small
  • All samples were injected from the same vial

ENVIRONMENT

  • 2695 Separations Module
  • 2489 UV/Vis Detector

CAUSE

Poor sensitivity of 2489 detector, 2695 not injecting the correct volume

FIX or WORKAROUND

  1. Power cycle the 2489 detector.
  2. If it does not improve, check the lamp energy of the 2489 detector (refer to the KCS article Noisy baseline on UV detectors - WKB933 for the procedures).
  3. Confirm the injection volume from vial weight
    - Add water into the vial.
    - Specify the method for normal measurement, set the injection volume to 50 µL, and run.
    - Measure vial weight before and after injection.
