SYMPTOMS
- The spectrum shape is significantly different compared to the spectrum shape of the same compound acquired by other PDA detectors

CAUSE
- The resolution setting of the instrument method is different
- The check boxes of "Interpolate the 2nd order filter region" and "Interpolate 656 nm" in the instrument method are OFF
- The PDA detector cell is dirty
FIX or WORKAROUND
- Check the resolution setting of the instrument method.
→ Is it equivalent to the value used for acquisition of the spectrum being compared? - Check if the "Interpolate the 2nd order filter region" and "Interpolate 656 nm" options of the instrument method are checked.
→ If it is OFF, turn it ON and run the measurement, and then compare the shapes. - Clean PDA detector cells.
→ If the cause is dirt on the cell, see if cleaning can help.
■ General cleaning
① Remove the column and connect to the detector with a union.
② Flush all mobile phase lines with 100% water at a flow rate of 0.2 mL/min for half a day.
③ Flush all mobile phase lines with 100% IPA at a flow rate of 0.2 mL/min for half a day.
④ Flush all mobile phase lines with 100% water at a flow rate of 0.2 mL/min for one hour.
⑤ Flush all mobile phase lines with 1% formic acid in water at a flow rate of 0.2 mL/min for three hours.
⑥ Flush all mobile phase lines with 2% ammonia in water at a flow rate of 0.2 mL/min for three hours.
⑦ Flush all mobile phase lines with 100% water at a flow rate of 0.2 mL/min for three hours.
*Use wet prime when replacing the solvent.
* In addition to the solvents listed above, if there is a solvent that is likely to easily dissolve the potential contamination (mainly the sample), flush with that solvent.
* The specified time is a reference.
