How to install a scrubber on an ACQUITY UPLC - WKB16133

OBJECTIVE or GOAL

Install a scrubber on an ACQUITY UPLC. When major contamination is observed coming from aqueous mobile phase, you can install a scrubber. This small chromatographic column absorbs contaminant, such as polyethylene glycol).

PROCEDURE

  1. Order the following parts:

186003107 - XBridge C18 5μm 2.1x20mm Column 

430002983 - ASSY, WELDED TUBE, SST, 6.0 LG, HP

  1. Ensure that the I-Class ACQUITY UPLC Binary Solvent Manager module has passed all operational tests, and prime solvent lines A1 and A2 with the best source of water.
  2. Perform a gradient test (Positive-ion polarity only) by running three replicates of Waters standard sample according to installation manual 715004586 (Xevo G2-XS), 715004250 (SYNAPT G2-Si), or 715005144 (Vion IMS QTof).

  3. Display the EIC chromatogram profile of sulfadimethoxine (m/z 311) for the three runs, calculate the average (n = 3) of pre-modification signal-to-noise value (RSM with no extra processing), and note the standard deviation of peak Retention Time (should be ≤ 0.047 minutes).
  4. Disconnect the stainless steel tubing (430001208) between Vent Valve port 2 and the tee/filter at the filter mixer assembly.

  5. Connect the stainless steel tubing (430001208) to the inlet port of the XBridge column (186003107 - C18 5μm 2.1x20mm).

  6. Set up a 0.5-mL/min flow rate with 100% of A1 through the XBridge column.

  7. Flush the XBridge column into a waste beaker for 10 minutes, and leak-check the new connection.

  8. Connect the 430002983 tubing to the outlet port of the column. Ensure that water comes out the other end of the 430002983 tubing, and leak-check the new connection.

  9. Connect the free end of the 430002983 tubing to the filter mixer assembly, and leak-check this connection.

  10. Perform a post-modification gradient test by running three replicates of Waters standard sample according to installation manual 715004586 (Xevo G2-XS), 715004250 (SYNAPT G2-Si), or 715005144 (Vion IMS QTof).

  11. Display the extracted ion chromatogram profile of sulfadimethoxine (m/z 311) for the three runs, calculate the average (n = 3) of post-modification signal-to-noise value (RSM with no extra processing), and note the standard deviation of peak Retention Time (should be ≤ 0.047 minutes).

  12. Modification is successful if:

  • Standard deviation of m/z 311 peak Retention Time is ≤ 0.047 minutes
  • Average (n= 3) of post-modification signal-to-noise value on m/z 311 peak is not significantly different or greater than the value calculated pre-modification

 

Maintenance of the XBridge column:

Note:    Waters recommends that you maintain the column regularly (every one or two weeks) to ensure scrubbing/cleaning efficiency and longer service life.

  1. Divert the LC flow to waste.
  2. Set up the LC in order to flush the analytical column with 100% organic solvent for 10 minutes.
  3. Stop the LC flow and remove the analytical column.
  4. Remove the XBridge column from the front of the BSM, and then fit it into the column compartment before flushing the XBridge column with 100% organic solvent at 1 mL/min for at least one hour.
  5. Fit the XBridge column back onto the front of the BSM on the aqueous solvent line.
