How to verify the response value with Investigate when analyzing with Find 2D Peaks - WKB86801

OBJECTIVE or GOAL

  • Analysis was performed with the Peak detection setting set to "Use 2D peaks” of the Default Quantify in Analysis Method.
    The XIC Area value is displayed in the response, but I want to perform the same analysis with Investigate to verify that the same area value is displayed

PROCEDURE

1. On the Review screen, note the "Observed m/z" of the target compound.


2. Note the following settings in Analysis Method.


2-1. Value of MS extraction region
Click Target by Mass in Targeted Screen Settings.
Note the value in the Manual field of Extract mass chromatograms.

2-2. Value of Smooth
Click Find 2D Peaks in Peak Processing Settings.
Click Smooth in the Task field.
Note the Smoothing algorithm, Half width and Iterations.

2-3. Integrate settings
Click Find 2D Peaks in Peak Processing Settings.
Click Integrate in the Task field.
Note the Chro peak width and Peak detection threshold.


3. Select the Investigate on the Analysis screen.

4. Select the sample for which you want to calculate the Area value.


5. Click the Select a Chromatogram icon in the upper right corner, select ▲ of 1.TOF MSE , and check the TOF MSe TIC.


6. Click Processing Options.


7. Select Extract mass chromatograms in the Operation field.
Enter the “Observed m/z” noted in step 1 in the Parameter field.
Enter the value noted in 2-1 into Tolerance.
Click the Start button.

 

8. Select Smooth chromatogram in the Operation field.
Enter the Smoothing algorithm, Half width and Iterations values noted in 2-2 into the parameter fields of Type, Half width, and Iterations.
Click the Start button.

 

9. Select Integrate in the Operation field.
Enter the Chro peak width and Peak detection threshold noted in 2-3 into the Peak width and Detection threshold field.
Click the Start button.

 

10. Click the Show or hide additional information icon in the upper right corner.
Confirm the Peak area value of the retention time for the target compound.
