TargetLynx calculates the wrong concentration for analytes - WKB23320

SYMPTOMS

  • The standard concentrations look good and the calibration curve looks good
  • Concentrations are calculated for the analytes, but for the same response in the standard the analyte is given a much higher concentration

CAUSE

A different amount of internal standard was used in the standards than in the analytes, and the TargetLynx method is using a fixed concentration for the internal standard. So it is expecting the same internal standard response for every sample, regardless of whether it is a sample or analyte.

FIX or WORKAROUND

Remake the samples (standards and analytes) with the same amount of internal standard.

OR

  1. Add a Conc column (for example, Conc B) to the sample list.
  2. Fill in the Internal Standard concentration for each sample in the sample list. 
  3. Open the TargetLynx method and go to the calibration properties page for the internal standard.
  4. Change the concentration of the standard level from fixed to Conc B, and save the method.
  5. Reprocess the samples.
