What is the process for an SPE loading study? - WKB75867

Article number: 75867

ENVIRONMENT

  • SPE loading study
  • Oasis cartridges

ANSWER

Connect two devices together (one on top of the other), or take the load and wash fractions from A and load on B.
Call the top "A" and the bottom "B"; load approximately 1.5 times the predicted loading capacity, followed by a wash step.
Separate the two devices and finish the extraction separately.

Run assay for results.

There are three possible outcomes:

  1. Nothing on B, everything on A. (You did not reach the loading capacity for a device. Double the amount and try again.)
  2. A and B have approximately equal amounts. (You overloaded both devices Cut the sample amount in half and try again.)
  3. A has more than B. (You overloaded A. B retained the rest.)

A/(A+B) x amount loaded = Maximum Capacity

Because many factors can lead to variation, work at  approximately 50% of maximum capacity.
