ANSWER

Histidine contains nucleophiles, which compromise rapid labeling with RapiFluor-MS.

ADDITIONAL INFORMATION

The presence of histidine in a sample consumes a substantial amount of the RapiFluor-MS reagent before the reagent fully reacts with the N-Glycans present in a sample. It is advised that you dilute amine or thiol concentrations in a sample to less than 0.1 mM in order to alleviate this type of issue. Some samples may require a buffer exchange prior to enzymatic deglycosylation in the GlycoWorks RapiFluor-MS workflow.
